摘要

The unique role of a new (SCS)Ni cofactor in lactate racemase (LarA) is examined in this computational study. The Ni-dependent cofactor could increase dehydrogenation barriers and particularly destabilize the NADH-like pyruvate intermediates, due to the lower electrophilicity of the cofactor and the ring strain in the intermediates. This intermediate destabilization by a cofactor in LarA, which can promote racemization, is uncommon in enzymatic catalysis.
